Regulation of multidrug resistance by microRNAs in anti-cancer therapy

چکیده
Multidrug resistance (MDR) remains a major clinical obstacle to successful cancer treatment. Although diverse mechanisms of MDR have been well elucidated, such as dysregulation of drugs transporters, defects of apoptosis and autophagy machinery, alterations of drug metabolism and drug targets, disrupti on of redox homeostasis, the exact mechanisms of MDR in a specific cancer patient and the cross-talk among these different mechanisms and how they are regulated are poorly understood. MicroRNAs (miRNAs) are a new class of small noncoding RNAs that could control the global activity of the cell by post-transcriptionally regulating a large variety of target genes and proteins expression. Accumulating evidence shows that miRNAs play a key regulatory role in MDR through modulating various drug resistant mechanisms mentioned above, thereby holding much promise for developing novel and more effective individualized therapies for cancer treatment. This review summarizes the various MDR mechanisms and mainly focuses on the role of miRNAs in regulating MDR in cancer treatment.

Molecular mechanisms involved in multidrug resistance in breast cancer therapy
Breast cancer is the most prevalent cancer in women. Chemotherapy is the main strategy in the treatment of this disease especially in the advanced form of the disease. Despite the recent progress in the development of new chemotherapy, the effectiveness of these drugs has dramatically reduced due to multidrug resistance. The phenotype of multidrug resistance (MDR) can occur through different me...

Background and Objective: Tamoxifen is the most commonly used treatment for the patients with breast cancer called ER +, which prevents the expression of genes that are effective in the growth and proliferation of cancer cells by estrogen. Resistant to Tamoxifen is a major clinical problem in breast cancer treatment. In recent studies, the role of microRNAs in tamoxifen resistance has been rais...

Background: Multidrug resistance (MDR) is known to defeat most chemotherapies as one of the main anticancer strategies. The role of overexpression/overactivation of ABC transporters, especially P-glycoprotein (P-gp), in the development of chemotherapy has long been demonstrated. Salinispora is a marine actinomycete genus known for the production of novel bioactive metabolites. Methods: In this...
متن کاملMicroRNAs as biomarkers associated with bladder cancer
Bladder cancer is the fifth most common cancer with significant morbidity and mortality. Recently, numerous studies demonstrated that microRNAs are emerging as diagnostic biomarkers for bladder cancer. Specific miRNA profiles have been identified for several samples from patients with bladder cancer. MicroRNAs are noncoding RNA molecules of approximately 23 nucleotides that play important roles...

Objective(s):Multidrug resistance (MDR) of cancer cells is a major obstacle to successful chemotherapy. Overexpression of breast cancer resistance protein (BCRP) is one of the major causes of MDR. In addition, it has been shown that PI3K/Akt signaling pathway involves in drug resistance. Therefore, we evaluated the effects of novel approaches including siRNA directed against BCRP and targeted t...
